Overexpression of RhoGDI, a novel predictor of distant metastasis, promotes cell proliferation and migration in hepatocellular carcinoma

RhoGDI(RhoGDP‐dissociationinhibitoralpha,orRhoGDIα)wasidentifiedasaregulatorofRhoGTPases,butitsroleincancerremainscontroversial.Inthisstudy,increasedexpressionofRhoGDIwasdetectedinhepatocellularcarcinoma(HCC)celllinesandtissueswithhighlymetastaticpotential.RhoGDIoverexpressioncorrelatedwithpostoperativedistantmetastasis.EnforcedexpressionofRhoGDIinHCCcellssignificantlyenhancedcellproliferationandmigration.Conversely,knockdownofRhoGDIcausedaninhibitionoftheaggressivephenotypesofHCCcells.Furthermore,RhoGDIup‐regulatedRho,butnotRac,andenhancedPI3K/AKTandMAPKpathwayactivity.OurfindingssuggestthatRhoGDIoverexpressionisapredictorofdistantmetastasisandplaysanimportantroleintheprogressionofHCC...
