OxidativestressisakeypathologicalfeatureofParkinson'sdisease(PD).GlutathioneS‐transferasepi(GSTP)isaneuroprotectiveantioxidantenzymeregulatedatthetranscriptionallevelbytheantioxidantmasterregulatornuclearfactor‐erythroid2‐relatedfactor2(Nrf2).Here,weshowforthefirsttimethatuponMPTP‐inducedoxidativestress,GSTPpotentiatesS‐glutathionylationofKelch‐likeECH‐associatedprotein1(Keap1),anendogenousrepressorofNrf2,invivo.S‐glutathionylationofKeap1leadstoNrf2activationandsubsequentlyincreasesexpressionofGSTP.ThispositivefeedbackregulatorylooprepresentsanovelmechanismbywhichGSTPelicitsantioxidantprotectioninthebrain...
