Cloning and functional characterization of TCRP1, a novel gene mediating resistance to cisplatin in an oral squamous cell carcinoma cell line

Toexplorethemechanismsofchemotherapyresistance,wepreviouslyestablishedamulti‐drugresistantcellline,Tca8113/Pingyangmycin(Tca8113/PYM)andidentifieddifferentialexpressioninknowngenesandESTsusingmicroarrayanalysis.FromamongthoseESTswehavenowidentifiedanovelgeneproducinganmRNAof1834nucleotidestranslatedintoaproteinhaving235aminoacids.Thisgenewasdenominatedastonguecancerresistance‐associatedprotein1gene(TCRP1,accessionnumber:EF363480).Wefurtherdetermineditsfunctionalcharacteristics.TheresultsdemonstratethatTCRP1mediatesaspecificresistancetocisplatininTca8113cellsbyreducingthecisplatin‐inducedapoptosis.ThissuggeststhatTCRP1mightbeanovelmoleculartargettodevelopagentstoreversecisplatin‐inducedchemoresistance...
