Central PGE2 exhibits anxiolytic‐like activity via EP1 and EP4 receptors in a manner dependent on serotonin 5‐HT1A, dopamine D1 and GABAA receptors

Wefoundthatcentrallyadministeredprostaglandin(PG)E2exhibitedanxiolytic‐likeactivityintheelevatedplus‐mazeandopenfieldtestinmice.AgonistsselectiveforEP1andEP4receptors,amongfourreceptorsubtypesforPGE2,mimickedtheanxiolytic‐likeactivityofPGE2.Theanxiolytic‐likeactivityofPGE2wasblockedbyanEP1orEP4antagonist,aswellasinEP4butnotEP1knockoutmice.CentralactivationofeitherEP1orEP4receptorsresultedinanxiolytic‐likeactivity.ThePGE2‐inducedanxiolytic‐likeactivitywasinhibitedbyantagonistsforserotonin5‐HT1A,dopamineD1andGABAAreceptors.Takentogether,PGE2exhibitsanxiolytic‐likeactivityviaEP1andEP4receptors,withdownstreaminvolvementof5‐HT1A,D1andGABAAreceptorsystems...
