To investigate the impact of increased osteoblastic activity on the proliferation and survival of MM plasma cells.Multiple myeloma (MM) is one of representative hematologic malignanciescausing the skeletal related events. Dysregulation of bone remodeling(increased osteoclastic activity and decreased osteoblastic activity) hasbeen known as a key pathomechanism of disease progression and skeletalrelated events. However, decreased proliferation of MM at fractured site hasbeen noted in clinical situation regardless of the systemic disease activity.To investigate effects of increased osteoblastic activity on survival andproliferation of MM plasma cells, co-culture under various conditions wasdone. MM plasma cells (Cell line: IM-9 from Korean cell line bank) werecultured in culture media (control) and co-cultured with human mesenchymal stem cells (hMSCs, SS16-P2 from Catholic institute of cell therapy,group I), osteoblasts (OBs) induced from hMSCs (group II) and bonemorphogenic protein-2 (BMP-2, group III). Proliferation (ERK and Ig Gexpression) and apoptosis (FACS with annexin V method, caspase-3 andstat-3 expression) of cultured MM plasma cells and expression of sclerostin were assessed.
