High ω3-polyunsaturated fatty acids in fat-1 mice prevent scopolamine-induced memory impairment through BDNF signaling

Polyunsaturated fatty acids (PUFAs) are critical for opti-mal brain health and are involved in psychiatric and neurologicalailments. Here, we show the effects of scopolamine on memoryimpairment in the ?-3 PUFAs-overexpressing fat-1 mouse hip-pocampus. We performed Y-maze and passive avoidance teststo evaluate the memory function of fat-1 mice after administer-ing scopolamine. Fat-1 mice showed induced alternation in theY-maze test and increased latency in the passive avoidance test.The effects of scopolamine on damaged hippocampal neurogen-esis were confirmed by an increase in Ki-67- and DCX-positivestained cells in the fat-1 mice. Western blot analysis revealedincreased expression of brain-derived neurotrophic factor (BDNF)and phosphorylated cAMP response element-binding (p-CREB)proteins.Weshowedthat?-3PUFAshadaprotectiveeffectagainstscopolamine-induced apoptosis based on expression of cleaved-caspase 3 protein. These findings indicate that, in response to ?-3PUFAs, scopolamine-treated fat-1 mice were protected from gran-ularcelllossandexhibitedincreasedBDNFsignalinganddecreasedapoptosissignaling.Theseprocessesmayunderliegranularcellsur-vival and provide potential therapeutic targets for treatment ofmemory impairment.
