BackgroundandPurpose:Detectionofmouseparvovirus1(MPV)dependsonuseofserologicandpolymerasechainreaction(PCR)assays.Theseassayswereevaluatedfortheirabilitytodetectvirus-specificantibodiesorviralDNAinmultiplestrainsandagesofmiceinoculatedwithMPV.brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Twelve-week-oldICR,BALB/c,C3H,C57BL/6,andDBA/2miceandfour-andeight-week-oldICRmicewereinoculatedwithMPV.Serumwasharvestedfourweeksafterinoculationandanalyzedbyuseofrecombinantnonstructuralprotein1(rNS1)enzyme-linkedimmunosorbentassay(ELISA),minutevirusofmice(MVM)ELISA,andMPVindirectfluorescentantibody(IFA),MVMIFA,andMPVhemagglutinationinhibition(HAI)assays.SelecttissueswereharvestedandanalyzedbyuseofanMPV-specificPCRassay.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResults:ThenumberofmiceineachgroupwithdetectableMPV-specificantibodiesorMPVDNAvariedwithmousestrain,mouseagewheninoculated,andviraldose.Seroconversioninmiceinoculatedat12weeksofagewasdetectedalmostexclusivelybyuseoftheMPVIFAandMPVHAIassays,whereasseroconversioninalmostallmiceinoculatedat4and8weeksofagewasdetectedbyuseofallimmunoassaysexcepttheMVMELISA.ViralDNAwasdetectedbyuseofPCRanalysisinallstrainsandagesofmiceexceptDBA/2mice.brxmlns="http://pub2web.metastore.ingenta.coms/"/brConclusions:MousestrainandagehaveimportantrolesinseroconversiontononstructuralandstructuralMPVantigensandpersistenceofviralDNAinmousetissues.Therefore,diagnosticserologictestingandPCRanalysisshouldbeconsideredwithinthecontextofmousestrainandageatthetimeofMPVexposure,especiallywhensentinelmiceareusedforsurveillance.
