Purpose:InastrainoftheLong-EvansCinnamon(LEC)rats,wefoundspontaneouslyhyperactiveanimalsdesignatedas#8220;wiggling,#8221;andestablishedacongenicwiggling(Wig)ratbytransferringthegenefromtheLECtotheWistarKingAptekman/Hokkaido(WKAH)strain.WeevaluatedthefeasibilityoftheWigratforananimalmodelofhumanattentiondeficithyperactivedisorder(ADHD).brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Modeofinheritancewasexaminedbyuseoflinkageanalyses.Motoractivity,behavior,andworkingmemorywereassessedbyuseofelectricdigitalcounters,openfieldtest,andY-mazeandwater-mazetests.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResults:Theabnormalbehavior,includinghyperactivity,wastransmittedinautosomalrecessivemode.Diurnalandnocturnalmotoractivityof12-to14-week-oldWigratswasmarkedlyhigherthanthatofcontrols,andthishyperactivitywasmoreprominentduringnighttimethandaytime.Ambulationintheopen-fieldtestwassignificantlyincreasedinWigrats,butrearingwasdecreasedinWigrats,comparedwithcontrols.ResultsoftheY-mazetestsindicatedthatspontaneousalternationbehaviorwassignificantlyimpairedinWigrats,althoughtherewasnodifferenceinthetotalarmentries.Thewater-mazetestcouldnotbeperformedbecause,whenexposedtowater,Wigratspanickedandalmostdrowned.brxmlns="http://pub2web.metastore.ingenta.coms/"/brConclusions:Wigratsarehyperactiveandhaveimpairedworkingmemoryandimpulsivebehavior,asassessedbythemotoractivityandopen-fieldtestsandtheY-mazetest,andtheseabnormalitiesaretransmittedbyasinglegenewithMendelianpattern.WigratsrepresentanexcellentanimalmodelofhumanADHD.
