Dietarydeficiencyof#969;3fattyacidduringdevelopmentleadstoimpairedcognitivefunction.However,theeffectsofmultiplegenerationsof#969;3fatty-aciddeficiencyoncognitiveimpairmentremainunclear.Inaddition,wesoughttotestthehypothesisthatthecognitiveimpairmentsof#969;3fatty-acid#8211;deficientmicearemediatedthroughthearachidonicacid#8211;cyclooxygenase(COX)pathway.Toaddresstheseissues,C57BL/6Jmicewerebredfor3generationsandfeddietseitherdeficient(DEF)orsufficient(SUF)in#969;3fattyacids.Atpostnatalday21,theF3offspringremainedonthedam'sdietorwereswitchedtotheoppositediet,creating4groups.Inaddition,2groupsthatremainedonthedam'sdietweretreatedwithaCOXinhibitor.At19wkofage,spatial-recognitionmemorywastestedonaY-maze.Resultsshowedthat16wkofSUFdietreversedthecognitiveimpairmentofF3DEFmice.However,16wkof#969;3fatty-acid#8211;deficientdietimpairedthecognitiveperformanceoftheF3SUFmice,whichdidnotdifferfromthatoftheF3DEFmice.Thesefindingssuggestthatthecognitivedeficitsaftermultigenerationalmaintenanceon#969;3fatty-acid#8211;deficientdietarenotanygreaterthanarethoseafterdeficiencyduringasinglegeneration.Inaddition,treatmentwithaCOXinhibitorpreventedspatial-recognitiondeficitsinF3DEFmice.Therefore,cognitiveimpairmentduetodietary#969;3fatty-aciddeficiencyappearstobemediatedbythearachidonicacid#8211;COXpathwayandcanbepreventedby16wkofdietaryrepletionwith#969;3fattyacidsorCOXinhibition.
