Thesucrosebreathtest(SBT)isasimplenoninvasivetechniqueusedcurrentlytodetermineintestinalabsorptivefunctioninhumansandrodents.However,todate,thetesthasnotbeenadaptedforuseinswine.Duringweaning,intestinalsucraseactivityinpigletstemporarilydeclinesinresponsetostressorsandiscommonlyusedasamarkeroftheintestinalresponsetoweaning.HereweassessedthesucrosedoseneededforusingtheSBTinpiglets.Sixrandomlyallocatedpigletswereorogastricallygavagedwithsup13/supC-labeledsucroseatadoseof2g/kg;breathsampleswerecollectedformeasurementofsup13/supCOsub2/subondays0(approximately17hafterweaning),5,and10afterweaning.TheresultantSBTvalue(cumulativedoseat90min)wasdecreasedby46%onday5afterweaningrelativetobaselinelevels,consistentwithtemporalchangesingastrointestinalsucraseactivityassociatedwithweaning.Weconcludethatasucrosedoseof2g/kgissatisfactorytoconductSBTstudiesinpiglets.Withfurtherdevelopment,theSBTmayprovideanewtooltononinvasivelymonitordigestivefunctioninweanedpiglets,toassesstheeffectsofnutritionalstrategiesonintestinalhealth,andasanindicatorofgutintegrityandfunctioninswinemodelsofhumangastrointestinaldisease.
