Interaction of H_2-receptor antagonists (cimetidine and ranitidine) and high blood alcohol concentrations. Experimental investigations

In einer randomisierten, doppelblind Placebo-kontrollierten Studie werden 12 Probanden an fünf Versuchstagen H_2-Rezeptor-Antagonisten (H_2RA: Cimetidin und Ranitidin) bzw. Placebo - jeweils mit und ohne Alkohol - postprandial appliziert. Die Dosierung betrug für Cimetidin 2 x 400 mg pro Tag/die für Ranitidin 2 x 150 mg/die, für Alkohol 1,4 g/kg Körpergewicht. Es werden die kinetischen Daten der H_2RA im Serum - mit und ohne Alkoholeinfluß - verglichen; es werden ferner die kinetischen Daten des Alkohols im Serum (BAK) - mit und ohne H_2RA-Einfluß - verglichen. In keinem Fall konnten Unterschiede festgestellt werden. Mittels psychomotorischer Testverfahren (Wiener Determinationsgerät, Wiener Reaktionsgerät) und Registration der subjektiven Befindlichkeit wird ferner der Einfluß von H_2RA - mit und ohne Alkohol-- erfaßt. Es können zwar die bekannten negativen Einflüsse des Alkohols nachgewiesen werden; es ergeben sich jedoch keine Hinweise auf negative oder positive Einflüsse zusätzlicher Art durch H_2RA.%In randomised double-blind, placebo-controlled study 12 volunteers were given the H_2 receptor antagonists (H_2RA) cimetidine and ranitidine - with and without alcohol - or placebo with alcohol - after meals on five days of the study. The dosage for cimetidine was 400 mg twice daily, for ranitidine 150 mg twice daily and for alcohol 1.4 g/kg body weight. The kinetic data of the H_2RA in the serum were compared with and without the effect of alcohol; furthermore, the kinetic data of the alcohol in the serum (BAC) were compared with and without the effect of H_2RA. No differences were found in any of the cases. The effects of H_2RA - with and without alcohol - were investigated by psychomotor testing (Vienna determination apparatus, Vienna reaction apparatus) and registration of the subjective mood. Although the known negative effects of alcohol were demonstrated, there were no indications of negative or positive effects by H_2RA.
