The Influence of Temperature on Epidermal Cell Migration in vitro

The effect of temperature on epidermal cell migration in vitro has been studied in guinea-pig skin. A total of 252 explants were cultured, 26 at each of several temperatures ranging from 27° to 45°. Half the explants were incubated in the presence of foetal calf serum and half in the presence of fresh guinea-pig serum: incubation times were 24, 48 or 72 hours. Explants were evaluated by examining stained paraffin sections and the degree of epidermal cell migration was scored according to an arbitrary scale. Raising the temperature was found to increase both the extent and the incidence of migration up to a temperature of 40°. Both types of serum used promoted migration although the results suggested that autologous serum was more effective.
