Objective: To investigate the effects of curcumin on the pathological alterations and tumor necrosis factor-alpha(TNF-α) of liver tissue in non-alcoholic fatty liver disease (NAFLD) rabbits. Methods:Thirty-four rab⁃bits model of NAFLD were randomly divided into 2 groups: control group(17 animals) and treatment group (17 an⁃imals).Both groups were fed standard diet.The treatment group received oral curcumin at dose of 200mg·kg-1·d-1 and the control group received equivalent oral curcumin-free sodium chloride solution once daily for 1 month. The expression of TNF-α protein was measured by immunohistochemistry and the pathological alterations in the liver were examined. Results: Curcumin significantly decreased the expression of TNF-α protein(P<0.05) and alle⁃viated the severity of inflammation and fibrosis in the liver sections. The expression of TNF-α protein was posi⁃tively correlated with scores of inflammation and fibrosis. Conclusion: Curcumin can improve the hepatic inflam⁃mation and hepatic fibrosis of NAFLD rabbits.% 目的:观察姜黄素对非酒精性脂肪肝病(NAFLD)家兔的肝脏病理学变化及肝组织TNF-α表达水平的影响.方法:复制成功的NAFLD模型家兔34只随机分为姜黄素组和模型组,各17只,均予基础饲料喂养,治疗组每天给予姜黄素200mg灌胃1次,模型组每天给予生理盐水灌胃1次,治疗1个月.免疫组织化学法分析TNF-α表达,观察肝组织病理学变化.结果:姜黄素组TNF-α表达较模型组显著降低(P<0.05),肝炎症活动及纤维化程度明显好转.姜黄素组TNF-α表达水平与肝炎症活动度、纤维化呈正相关(P<0.05).结论:姜黄素可有效改善肝脏炎症活动及纤维化程度,从而防治NAFLD.
