大肠癌围手术期气虚证与阴虚证患者基因差异表达图谱研究

         

摘要

目的:研究大肠癌围手术期气虚证与阴虚证患者的特征性基因差异表达谱。方法大肠癌手术患者8例,以围手术期中医辨证,气虚证4例(气虚证组),阴虚证4例(阴虚证组),采集8例患者大肠癌组织及瘤周正常组织,采用含有45033个己知基因的人全基因组芯片技术检测大肠组织内差异基因表达情况,比较各组的差异表达基因,利用生物分子功能注释系统GO和pathway对相关基因进行生物信息学分析。结果大肠癌围手术期气虚证瘤体组和瘤周正常组差异表达基因共5044条,表达水平上调3400条,下调1644条;气虚证瘤体组与阴虚证瘤体组差异表达基因共1335条,表达水平上调547条,下调788条。经检索互联网生物学公共数据库GeneBank,获得每条基因的名称和其他基本信息。结论大肠癌围手术期气虚证组与瘤周正常组比较,存在的组织基因差异表达特征图谱,主要与细胞凋亡、细胞周期、血管平滑肌收缩、嘌呤嘧啶代谢、细胞信号传导、DNA复制等基因相关;气虚证组与阴虚证组瘤体比较,也存在组织基因差异表达特征图谱,主要是与炎症、免疫应答、矿物质吸收、细胞内吞、细胞黏附、遗传信息过程、肌动蛋白细胞骨架的调控、氮代谢、细胞信号传导等基因相关。%Objective Comparison of differently expressed genes between patients with Qi deficiency and Yin deficiency in perioperative period of colorectal carcinoma. Methods Eight patients with colon carcinoma undergone colectomy were divided into two groups according to the differential diagnosis with TCM syndrome during the peri-operative period: Qi deficiency group (n=4) and Yin deficiency group (n=4). The tumor tissue and corresponding non-tumor normal tissues were collected for detection of gene expression by human gene technology with 45 033 known human genes. The gene chip images of two groups were analyzed by Pathway and GO system. Results A total of 5044 genes expressed differently in colorectal tissue and corresponding non-tumor normal tissue in Qi defi-ciency group, among which 3400 were up-regulated and 1644 were down-regulated; the expression of 1335 genes were different between Qi deficiency group and Yin deficiency group, among which the expression was elevated in 547 genes and was decreased in 788 genes. After retrieving the public databank, the names and basic information of all these differently expressed genes were obtained. Conclusion Comparing tumor tissue with corresponding non-tumor normal tissue in Qi deficiency group, the differently expressed gene map included apoptosis, cell cycle, vascular smooth muscle contraction, purine and pyrimidine metabolism, signaling pathway, and DNA replication. The difference in gene expression between Qi deficiency and Yin deficiency group included inflammation, immune re-sponse, mineral absorption, endocytosis, cell adhesion, genetic information pathway, regulation of actin cytoskeleton, nitrogen metabolism, signaling pathway.
