当归对胶原性关节炎模型大鼠血清MCP-1、MMP-3的影响

     

摘要

Objective To investigate the effect of Chinese angelica on the expression of serum monocyte chemotactic protein 1 (MCP-1) and matrix metalloproteinase-3 (MMP-3) in collagen-induced arthritis (CIA) model rats. Methods Thirty Wistar rats were randomly divided into blank control group, CIA model group, methotrexate group, and low and high doses of Chinese angelica groups, with 6 rats in each group. Except for blank control group, CIA models were established in other four groups. Methotrexate group received methorexate in-traperitoneally twice a week, 1mg/kg. Chinese angelica decoction was given to rats by gavage at dose of 0.75g(kg· d) and 1.5g(kg·d) in low and high doses of Chinese angelica groups, respectively. Blank control group and model group were administered with equivalent volume of saline. The treatment course lasted for 3 weeks. The serum lev-els of MCP-1 and MMP-3 were observed and the swelling scores of rats' joints were calculated. Results The serum MCP-1 and MMP-3 increased in all CIA rats (P<0.05). Compared with model group, methotrexate group and high dose of Ch-inese angelica group had decreased serum MCP-1 (139.20±11.69pg/mL, 151.64±13.42pg/mL vs 179.21 ±13.79pg/mL, P<0.05) and serum MMP-3 (1.57 ±0.19ng/mL, 1.69 ±0.21ng/mL vs 2.04 ±0.27ng/mL, P<0.05), and lower swelling score of the joints (2.83±0.41, 3.33±0.52 vs 3.83±0.41, P<0.05). Conclusion Chinese angelica can reduce the serum level of MCP-1 and MMP-3 and alleviate the swelling of the joints in rats with CIA.%目的:观察当归对胶原性关节炎(CIA)模型大鼠血清单核细胞趋化因子-1(MCP-1)、基质金属蛋白酶-3(MMP-3)的影响。方法清洁级Wistar大鼠30只,随机分为空白对照组、胶原性关节炎(CIA)模型组、甲氨蝶呤组和当归低、高剂量组,每组6只。除空白对照组外,采用Ⅱ型胶原免疫法制备胶原性关节炎模型。、甲氨蝶呤组每次给药1mg/kg,每周2次,腹腔注射;当归低、高剂量组分别灌饲当归煎液0.75g(kg·d)和1.5g(kg·d),空白对照组和模型组灌饲等量生理盐水,持续3周。观察大鼠血清MCP-1、MMP-3及大鼠关节肿胀指数。结果造模后,各药物组大鼠血清MCP-1、MMP-3均明显升高(P<0.05)。与CIA模型组比较,甲氨蝶呤组、当归高剂量组大鼠MCP-1均有降低[(139.20±11.69)pg/mL、(151.64±13.42)pg/mL比(179.21±13.79)pg/mL,P<0.05];甲氨蝶呤组、当归高剂量组大鼠MMP-3均有降低[(1.57±0.19)ng/mL、(1.69±0.21)ng/mL比(2.04±0.27)ng/mL,P<0.05];甲氨蝶呤组、当归高剂量组的大鼠关节肿胀评分均有降低[(2.83±0.41)分、(3.33±0.52)分比(3.83±0.41)分,P<0.05)]。结论当归能不同程度降低胶原性关节炎大鼠血清MCP-1、MMP-3含量,且同时减轻大鼠关节肿胀指数。
