补中益气汤对脾虚慢传输型便秘小鼠结肠组织血管活性肠肽的影响

         

摘要

目的:观察补中益气汤对脾虚慢传输型便秘(STC)小鼠结肠组织血管活性肠肽(VIP)的影响,探对其治疗脾虚慢传输便秘的作用机制。方法32只雄性昆明小鼠随机分为正常组(8只)和造模组(24只),造模组采用番泻叶+限制饮水+控制饮食的方法复制小鼠脾虚STC模型,模型复制成功后,将造模组小鼠随机分为模型组、中药组(补中益气汤)和西药组(莫沙必利),各组灌胃对应的药物,1天1次,共7天,末次给药后检测各组小鼠粪便含水率、肠道推进率及结肠组织VIP含量。结果中药组小鼠粪便含水率(55.55±10.90)%,肠道推进率(69.84±9.67)%,分别高于模型组的(45.50±13.04)%和(50.44±14.45)%(P均<0.05),中药组小鼠结肠组织VIP水平(156.73±11.41)ng/L,低于模型组的(245.12±12.43)ng/L(P<0.05)。西药组小鼠粪便含水率、肠道推进率分别为(52.83±12.43)%,(65.15±13.56)%,与中药组比较差异无统计学意义(P>0.05),西药组小鼠结肠组织VIP水平(120.91±17.65)ng/L较中药组降低,差异有统计学意义(P<0.05)。结论补中益气汤能提高小鼠粪便含水率及肠道推动率,改善脾虚慢传输型便秘症状,其作用机制可能与降低结肠组织VIP水平有关。%Objective To investigate the effects of buzhongyiqi decoction on vasoactive intestinal peptide (VIP) in slow transit constipation (STC) mice with spleen deficiency, in an attempt to explore the underlying mechanism. Methods Thirty-two male Kunming mice were randomly divided into normal group (n=8) and model group (n=24). The model group was copied to STC model with spleen deficiency by folium sennae+limited water+control diet. Then, the model group were randomly divided into model control group, Chinese herb group (buzhongyiqi decoc-tion) and western medicine group (mosapride). Each group was treated one time a day by gavage with its corre-sponding drug, the course lasted for 7d. After the last treatment, the moisture content of feces, intestinal propulsion rates and the content of VIP in colonic tissue were detected. Results Moisture content of feces and intestinal propulsion rate in Chinese herb group were (55.5±10.90)% and (69.84±9.67)%, respectively, which were higher than those in model control group [(45.50±13.04)% and (50.44±14.45)%, P<0.05]. VIP level of Chinese herb group was lower than that of model control group [(156.73±11.41)ng/L vs (245.12±12.43)ng/L, P<0.05]. The moisture co-ntent of feces and Intestinal propulsion rate in western medicine group were (52.83±12.43)% and (65.15±13.56)%;no significant difference was found between Chinese herb group and western medicine group (P>0.05). The VIP level of western medicine group [(120.91±17.65)ng/L] was significantly lower than that of Chinese herb group (P<0.05). Conclusion Buzhongyiqi decoction can increase the moisture content of feces and intestinal propulsion rate and improve symptoms of STC with spleen deficiency. The underlying mechanism might be related to the decrease of VIP in colonic tissue.
