高含量全反式番茄红素的全合成方法研究

     

摘要

设计了番茄红素的全合成绿色新工艺,采用弱碱性交换树脂催化醛酮缩合,用格氏试剂诱导制备六碳醇,与亚磷酸三乙酯发生Wittig-Horner反应制备六碳膦酸酯,再以甲基庚烯酮为起始原料,经Darzen反应与六碳膦酸酯缩合,经过格氏,催化氢化,脱水,手性拆分制备全反式十五碳磷酸酯,与十碳双醛对接,得高含量全反式番茄红素。%A total synthesis of lycopene green production process was developed. The synthetic steps were as follows: the condensation between formaldehyde and acetone was catalyzed by weak-base exchange resin, and then the hexacarbinal was prepared by introducing Grignard reagents. The 3-methyl-2-penten-4-yn-1-phosphonate was prepared via the Wittig-Horner reaction between phosphorous acid triethyl ester and hexacarbinal. The all-trans 3,7,11-trymethyldodeca-2,4,6,10- tetraenylphosphonate was produced from methyl heptenone and 3-methyl-2-penten-4-yn-1-phosphonate via the Darzen reaction, Grignard reaction, catalytic hydrogenation, dehydration and enantioseparation. Finally, the target product high levels and all-trans lycopene was obtained by the reaction between all-trans 3,7,11-trymethyldodeca-2,4,6,10-te-traenylphosphonate and 2,7-dimethyl-2,4,6-octatriene-1,8-dial.
