目的:选用大孔树脂纯化苦豆子籽提取液的生物碱的纯度,提高生物碱的纯度。方法:pH=9,浓度为2.0~2.5 mg/mL的料液,抽滤加氯化钠(1.0 mol/L),以树脂重量与样品的体积比为1∶3上柱,用3倍样品体积pH=9的30%乙醇先洗脱,再用3倍样品体积pH=9为70%乙醇洗脱。结论:选用非极性D101大孔树脂直接从苦豆子籽提取液中分离纯化生物终产品总碱纯度可达60%以上,洗脱率为97%以上,分离效果好。%Objective:To increase the purity of alkaloids by applying macroporous resin to total alkaloids from the extract of KuDouZi (Sophora alopecuroides L) seeds. Methods:Feed liquid, scaling pH=9, in the concentrations of 2.0 and 2.5 mg/mL, leaching and adding sodium chloride (1.0 mol/L), the weight of macroporous resin and the volume of the samples were in the ratio of 1:3 on the column, three times of the volume of the samples, pH=9, were used to elute firstly, and then three times volume of 70%ethanol with pH=9 were used. Conclusion:Non-polar D101 macroporous resin is chosen to separate and purify total alkaloids of the extract of KuDouZi seeds directly, which could make the purity of total alkaloids more than 60%, elution rate higher than 97%and obtaining good separation effects.
