Objective:To investigate the relationship between adiponectin gene SNP +45 (T/G) polymorphism and 8-isoprostaglandin F2α, a index of oxidative stress in type 2 diabetes (T2DM). Methods: The SNP +45 (T/G) polymorphism was detected by PCR-RFLP in 120 T2DM patients. The serum concentration of 8-isoprostaglandin F2α was measured by enzyme-linked immunosorbent assay method. The control group was selected from healthy people. Results: The serum level of 8-isoprostaglandin F2α was higher in T2DM group than that of control group. The frequency of adiponectin SNP45 site of GG was higher in T2DM group than that of control group (P < 0.05). In T2DM group the value of body mass index (BMI) was higher in GG genotype compared with that of TT and TG genotypes (P < 0.05). There were no significant differences in the serum concentration of 8-isoprostaglandin Fa, between TT, TG and GG genotypes (P > 0.05). Conclusion: There might be no significant relevance between adiponectin gene SNP +45 (T/G) polymorphism and oxidative stress in type 2 diabetes.%探讨脂联素基因SNP+45(T/G)多态性与2型糖尿病(T2DM)氧化应激指标8-异前列腺素F2a之间的关系.方法:聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)方法检测120例T2DM患者(T2DM组)脂联素SNP+45(T/G)多态性,同时选取正常体检者60例为对照组,采用竞争酶免疫测定的方法检测T2DM患者血清8-异前列腺素F2a水平,检测各组空腹血糖、血脂及空腹胰岛素水平,计算HOMA-IR等指标.结果:T2DM患者血清8-异前列腺素F2a水平高于对照组,T2DM患者脂联素SNP+45位点GG基因型频率高于对照组(P<0.05).T2DM组内GG型的体质量指数(BMI)高于TT及TG型(P<0.05);TT、TG及GG基因型血清8-异前列腺素F2a水平差异无统计学意义(P>0.05).结论:脂联素SNP+45(T/G)多态性与T2DM氧化应激之间可能无相关关系.
