粉尘螨抗原对脐血Th1/Th2细胞因子表达的影响

         

摘要

目的:观察新生儿脐血单个核细胞(CBMCs) Th1/Th2细胞因子的表达,探讨家族过敏史和过敏原刺激对健康足月新生儿Th1/Th2细胞偏移的影响.方法 健康足月新生儿30名,家族过敏史阳性组(AD组)及阴性组(对照组)各15名,分离CBMCs,经不同浓度的粉尘螨刺激后,用ELISA法检测培养上清中白细胞介素-4(IL-4)和γ-干扰素(IFN-γ)的含量.结果 (1)低浓度粉尘螨刺激后,与对照组相比,AD组IL-4显著增高,IFN-γ显著降低(均P<0.01);(2)高浓度粉尘螨刺激后,与对照组相比,AD组IL-4显著增高(P<0.01),但IFN-γ无明显变化(P>0.05).(3)对两组新生儿随访观察1年,AD组过敏性疾病发生率明显高于对照组(P<0.05).结论 母亲有家族过敏史的新生儿,CBMCs呈现Th2相对优势,提示家族过敏史对新生儿Th1/Th2平衡存在影响.%Objective To show the effects of family allergic history and allergen stimulation over the excursion of healthy neonate Thl/Th2 cell, by testing the expression of CBMCs Thl/Th2 cytokines. Methods Thirty healthy newborn babies were divided into two groups, one was composed of the babies with family allergic history as the testing group (AD group) ; the other was composed of the babies with no family allergic history as the control group. CBMCs were isolated by Ficoll gradient centrifugation and were stimulated by different concentration of Dermatophagoides farinae(Der f) for 48 h. The concentrations of IFN-γ and IL-4 in the culture supernatant were determined by ELISA assay. Results (1) Compared with the control group, the concentration of IL-4 in the culture supernatant was significantly increased and the concentration of IFN-7 was significantly decreased in low density Der f stimulated group(P<0.01). (2) In high density Der f stimulated group, the concentration of IL-4 was significantly increased than that in the control group (P <0. 01 ). However, there was no significant difference in the concentration of IFN-7 in the testing group there and the control group (P >0.05). (3) Allergic diseases happened significantly more in children with family allergic history than those in the families allergic history in one year period of follow-up uin the two groups. Conclusion In the group of healthy newborn babies with family allergic history, secretion of IL-4 increases significantly which means Th2 cells are dominant. It impliesthat family allergic history has effect on the balance of neonate Th1/Th2 cells.
