鸳鸯茉莉花瓣蛋白质双向电泳技术的优化

             

摘要

In order to find the method of two-dimensional electrophoresis technology in Brunfelsia acuminate petals, phenol extrac-tion method was adopted to optimize the sample volume and IPG strips and protein crucial steps. The results showed that protein iso-lation of B.acuminate petals was better in the range of pH 4.0-7.0, 24 cm IPG strip in the protein electrophoresis figure. When the sample volume was 1.5 mg, large and clear spots were observed, and a higher resolution in the 2-DE patterns was obtained.%为探索适用于鸳鸯茉莉花瓣蛋白质双向电泳技术的方法,采用酚抽提取法对IPG胶条及蛋白质上样量等关键步骤进行优化. 结果显示,鸳鸯茉莉花瓣蛋白质在pH值为4.0-7.0,24 cm IPG胶条范围分离效果较好,并且当上样量为1.5 mg时,在双向电泳图谱上检测到的蛋白点较多且清晰,分辨率较好.
