茵陈蒿汤对酒精性诱导脂肪肝大鼠的护肝、抗自由基损伤和降血脂作用

摘要

目的：探讨茵陈蒿汤对酒精性脂肪肝大鼠自由基损伤、血脂及肝脏结构和功能的作用。方法以10％酒精加高营养饲料喂饲6W，造成大鼠酒精性脂肪肝模型，分组给药；口服不同剂量茵陈蒿汤或水飞蓟宾100mg・ kg－1，给药4W和继续服10％酒精4W后，取血清测 AST、ALT、ALP活性和TC、TG、HDL－C、LDL－C含量，以及计算动脉硬化指数（ AI）；处死大鼠，取肝组织做病理学检查。结果茵陈蒿汤能抑制病鼠血清AST、ALT活性（P＜0．01），提高HDL－C含量（P＜0．01），AI下降（P＜0．01）；肝组织病理检查，茵陈蒿汤与水飞蓟宾均能明显改善肝组织水样和脂肪变性，差异有显著性（P＜0．05～0．01）。结论茵陈蒿汤能对抗大鼠酒精性脂肪肝，其机制与对抗酒精肝毒性和减少肝脏脂肪沉积有关。%OBJECTIVE TO study he effects of Yinchenhao decotion( YCHD) on liver structure and function, free Radical Injury and blood lipid in rats with alcohol fatty liver.METHODS To observe the effects of YCHD( in-tragastric administration,for qd ×4 weeks) on serum activities of alanine aminotransferase (ALT),aspartate amin-otransferase(AST),alkaline phosphatase(ALP)and serum contents of total cholesterol(TC),triglyceride(TG) high density lipoprotein cholesterol(HDL-C) and low density lipoprotein cholesterol(LDL-C),and calculate Arterio-sclerosis index( AI) ,malondialdehyde ( MDA) ,ectivity of superoxide dismutase( SOD) in the hepatic tissue and he-patic histopathology observed by optical microscopy,which the rats with alcoholic fatty liver were fed by high lipid food and drunk 10%alcohol qd ×10 week.RESULTS YCHD protecting effects in the rats were significant that se-rum activities of AST,ALT were decreased(P<0.05~0.01),the levels of TG,LDL-C and atherosclerotic index (AI)were reduced(P<0.01) and content of HDL-C was enhanced(P<0.01),in the hepatic tissue concentra-tions of MDA were lowered(P<0.01) and hydropic and fatty degeneration of hepatocytes were restored(P<0.01), as compared with the model control by ethanol-treated rats with alcohol fatty liver.CONCLUSION YCHD can resistance alcoholic fatty liver in rats,pharmacological mechanisms comprise anti-oxidation and removing free radi-cal,inhibiting lipid per-oxidation;adapting blood lipid component and reducing fatty sediment in hepatocytes and hlood vessel.
