UPLC－MS法测定灵芪养心口服液中黄芪甲苷的含量

摘要

目的：建立灵芪养心口服液中黄芪甲苷的含量测定的新方法。方法采用ACQUITY UPLC BEH C18色谱柱，以乙腈－水（35∶65）为流动相。在ESI正离子模式下，选择m／z 807．5离子进行 SIR检测。结果方法检出限0．2μg・ mL －1，黄芪甲苷线性范围为0．1026～10．26μg・mL －1（r＝0．9990），加标回收率及其RSD为97．96％（RSD％＝1．26）。结论该方法灵敏度高、稳定性好和快速的特点，可以作为灵芪养心口服液的质量控制标准。%ABSTRACT:OBJECTIVE To establish a UPLC-MS method for determinating astragaloside Ⅳ in oral liquid of Lingqi Yangxin.METHODS A ACQUITY UPLC BEH C18 column(2.1 ×50mm,1.7μm).and a mixture of aceto-nitrile-water(35∶65) as the mobile phase was adopted.The analytes were detected by ESI-MS in SIR mode with m/z 807.5 as the selected ion.RESULTS The detection limit of astragaloside Ⅳwas 0.2μg・ mL-1 .the linear range was 0.1026~10.26μg・ mL-1 ( r =0.9990 ) .The average recovery ( n =6 ) was 97.96%( RSD% =1.26 ) .CONCLUSION The proposed method has the characteristics of high sensitivity ,good stability and high speed .It can be used for quality control in the production of oral liquid of Lingqi Yangxin .
