The high resolution 2-DE maps of total proteins from roots, stems and leaves in maize were obtained through improved TCA/acetone method. 1100 ±89, 1275 ±72 and 1327 ±98 spots were detected on the 2-DE maps (pI 4-7) of roots, stems and leaves in maize respectively. The distribution of protein spots on 2-DE maps was intensively located in the range of molecular weight ( MW) 25-75 kD and pI 5-6. The impurities of protein samples could be removed effectively after the sample cleaning procedures were optimized and aqueous acetone was used for the secondary precipitation. This study provided a more convenient and applicable method for maize proteomic analysis using 2-DE.%利用改进的三氯乙酸/丙酮法对5叶期玉米根、茎、叶的总蛋白质进行双向电泳获得了高分辨率的电泳图谱.在等电点4~7范围内,根、茎、叶电泳图谱上的蛋白质点数分别为1100±89、1275±72和1327±98个.3个器官蛋白质点的分布在分子量25~75 kD和等电点5~6的范围击相对集中.通过优化样品的清洗程序结合对蛋白质在丙酮水化液中的2次沉淀能够较好地除去样品中的杂质而保证电泳质量.优化后的方法可用于玉米幼苗的根、茎、叶总蛋白质双向电泳的试验研究.
