为了阐明河流型和沼泽塑水牛DGAT1基因的第8外曼子的群体遗传差异和特征,采用PCR-SSCP和PCR产物直接测序技术对11个水牛群体402份样品进行了群体变异检测,并结合GenBank数据库上已发表的牛科物种DGAT1基因序列进行了比较分析.结果显示,河流型和沼泽型水牛DGAT1基因第8外显子序列一致,序列全长为75 bp;与普通奶牛序列相比,河流塑和沼泽塑水牛DGAT1基因第8外显子都为X等位基因,揭示其与水牛的高乳脂宰、低泌乳量有关.序列比对发现K等位基因在牛科物种中普遗存在.本研究结果揭示了水牛中DGAT1基因第8外显子的遗传特征,可为其奶用性状选育利用提供遗传背最资料.%In order to make clear the genetic differences and population characteristics about exon 8 of DGAT1 gene, genetic variation of 402 samples from 11 buffalo populations were assayed by employing PCR-SSCP and FCR direct sequencing method. The sequences of DCAT1 of the bovine species in GenBank database were pooled together, the comparative genomies analysis was conducted. The results showed that the sequences of DGAT1 exon8 of river buffalo were identical with swamp buffalo, their length was 75 bp. Compared with the sequence of Holsteins cow, the sequences of DGAT1 exon8 in swamp and river buffalo were allele K, which indicate that it was associated with high milk fat percentage and low milk yield in buffalo. After sequence alignment, were allele K was universal existence in bovine species. The genetic characteristics of DGAT1 exon 8 disclosed in this study could provide genetic background information for the selection and utilization of milk traits in buffalo breeding.
