不同苎麻品种根际土壤细菌多样性

         

摘要

[Objective] In this paper,the changes in soil bacterial community composition and the diversity in rhizosphere soil of different ramie varieties were studied.[Method]The technology of high-throughput sequencing on Illumina Miseq platform combined with related bioinformatics analysis was adopted to investigate the rhizosphere soil bacteria on the deep root,the middle root type and the shallow root type ramie.[Result] 257 100 reads of 230 664 OTUs (operational taxonomic unit) with an average length of 420 bp were generated at 3 % cut off level under all treatments.Venn chart showed that the number of common bacteria OTUs between the different treatments were 459,and the number OTUs of bacterial groups between the deep root ramie and middle root ramie was the largest.Chao index of deep root ramie was significantly higher than that of shallow root type ramie (13.86 %),and the difference was not significant between deep root ramie and medium root ramie.Shannon index of the deep root ramie was significantly higher than that of the middle root ramie,and the difference was not significant between deep root type ramie and shallow root type ramie.Simpson index between the three treatments was not significant.Observed species index,Chao index,Shannon index,Simpson index of the deep root type ramie were higher than that of other two treatments,which indicated that soil bacterial population richness,soil bacteria superiority of the deep root type ramie higher than that of other two treatments.The diversity index of soil bacteria was correlated with organic carbon and pH value and positively correlated with available potassium.The dominant groups of phylum(relative abundance > 10 %),class(relative abundance > 10 %) and genus(relative abundance > 1%) were basically the same under the three treatments.The dominant groups of phylum were Bacteroidetes,Firmicutes and Proteobacteria,the dominant groups of class were Bacteroidia,Clostridia and Bacilli,and the dominant groups of genus had 18 under the three treatments.[Conclusion] (i) Compared with the middle root type and the shallow root type ramie,the rhizosphere soil bacteria in the deep root ramie was the most abundant.(ii)The bacterial diversity in ramie rhizosphere soil was highly correlated with soil organic carbon and pH and positively correlated with available potassium.(iii) The dominant rhizosphere bacteria in deep rooted,middle rooted and shallow rooted ramie soils were basically the same.%[目的]本文研究了不同苎麻品种根际土壤细菌群落组成和多样性的变化.[方法]利用Illumina平台Miseq高通量测序技术结合相关生物信息学对深根型、中根型、浅根型苎麻根际土壤细菌进行高通量测序及分析.[结果]测序共获得230 664个OTUs,257 100条读数,平均读长420 bp.Venn图表明不同处理之间共有的细菌OTU数目为459个,深根型苎麻与中根型苎麻共有的细菌类群最多.深根型苎麻Chao指数显著高于浅根型苎麻13.86%,与中根型苎麻差异不显著.深根型苎麻Shannon指数显著高于中根型苎麻,与浅根型苎麻差异不显著.3个处理间的Simpson指数差异不明显.Observed species指数、Chao指数、Shannon指数、Simpson指数总体上呈现深根型苎麻高于其他2个处理,说明深根型处理土壤细菌种群丰富度、土壤细菌优势度高于其他2种处理.苎麻根际土壤细菌多样指数与土壤有机碳、pH值相关性较高,与速效钾呈正相关关系.3个处理的土壤根际细菌优势门(相对丰度>10%)、优势纲(相对丰度>10%)、优势属(相对丰度>1%)都基本相同.3个处理的土壤细菌优势门有拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)和变形菌门(Proteobacteria).3个处理的优势纲有拟杆菌纲(Bacteroidia),梭菌纲(Clostridia),芽孢杆菌纲(Bacilli).3个处理的优势属有18个.[结论]①与中根型、浅根型苎麻相比深根型苎麻根际土壤细菌最为丰富;②苎麻根际土壤细菌多样性与土壤有机碳、pH值相关性较高,与速效钾呈正相关关系;(③深根型、中根型、浅根型苎麻土壤根际细菌优势类群基本相同.
