小檗碱对高糖培养的背根神经节中疼痛相关因子的影响

             

摘要

Objective To study the effects of berberine on factors related to pain :P2X3 ,TRPV4 ,nNOS in dorsal root ganglia(DRG) cultured in high glucose environment .Methods Primary cultured DRG neurons were taken from new‐born SD rats .And the expression of factors in DRG neurons were detected by immunohistochem‐istry (IHC) and Western blotting .Results P2X3 ,TRPV4 and nNOS express in DRG neurons in normal condi‐tions ,and the positive neurons increased after cultured in high glucose ,and then they were inhibited by berberine which was detected by immunohistochemistry .The proteins of P2X3 and nNOS in high glucose environment were higher than those of control (P<0 .01 ,P<0 .01 vs control) and all of the three factors decreased after treated by berberine (P< 0 .01 ,P< 0 .05 ,P< 0 .01 vs high glucose) .Conclusion Berberine can inhibit the expression of P2X3 ,TRPV4 and nNOS in high glucose environment ,which will protect the DRG neurons from damage of dia‐betes mellitus ,and maybe the new method to improve painful diabetic neurpathy (PDN ) .%目的:探讨小檗碱对痛性糖尿病神经病变疼痛相关蛋白 P2X3、T RPV4、神经型一氧化氮合酶(nNOS )表达的影响及意义。方法新生大鼠离体培养背根神经节神经元,免疫组织化学法、蛋白质印迹法检测细胞表达阳性率及蛋白表达。结果 P2X3、T RPV4、nNOS正常条件下在背根神经节中均有表达。免疫组织化学法检测高糖培养条件下 P2X3、T RPV4、nNOS可看到阳性表达神经元增多,受体表达升高。小檗碱处理后, P2X3、T RPV4、nNOS表达均下降。蛋白质印迹法检测结果显示高糖条件下 P2X3、nNOS表达高于对照组( P<0.01),小檗碱处理后,P2X3、TRPV4、nNOS均较高糖组降低(P<0.01,P<0.05,P<0.01)。结论小檗碱可降低背根神经节神经元高糖环境下P2X3、TRPV4、nNOS的高表达,对糖尿病受损神经元有保护作用,可能为改善痛性糖尿病神经病变提供新途径。
