生物钟基因在腹部恶性肿瘤组织中的表达差异

         

摘要

Objective To explore the expression differences of clock genes in the abdominal malignant tumors.Methods The RNA-Seq expression of 14 kinds of clock genes in six abdominal malignant tumors and adjacent normal tissues (liver, gastric, pancreatic, kidney, rectal, and colon cancer) from the Cancer Genome Atlas (TCGA) database was integrated and evaluated to observe the differential expression of clock genes.We used the software edgeR in Bioconductor to standardize the RNA-Seq data.ResultsFourteen clock genes were significantly lowly expressed in the cancer tissues of six abdominal malignant tumors.Compared with the adjacent normal tissues, the expression levels of Cry2, CLOCK, Timeless, NPAS2, NR1D1, DEC1-2, and RORA were significantly lower in the liver cancer tissues (all P<0.05).Compared with the adjacent normal tissues, the expression levels of Per1-2, Cry1-2, CLOCK, Timeless, NPAS2, NR1D1-2, DEC2, and RORA were significantly lower in the kidney cancer tissues (all P<0.05).Compared with adjacent normal tissues, the expression levels of Per1-3, Cry1-2, Timeless, BMAL1, NR1D2, DEC1-2, and RORA were significantly lower in the colon cancer tissues (all P<0.05).Compared with adjacent normal tissues, the expression levels of Per1, Per3, Cry1-2, CLOCK, BMAL1, NPAS2, NR1D1, DEC2, and RORA were significantly lower in rectal cancer tissues (all P<0.05).Compared with the adjacent normal tissues, the expression levels of Per1, Per3, Cry2, CLOCK, Timeless, BMAL1, and NPAS2 were significantly lower in the gastric cancer tissues (all P<0.05).ConclusionDifferent clock genes are expressed differently in different abdominal malignant tumors.%目的 探讨生物钟基因在腹部恶性肿瘤组织中的表达差异.方法 利用癌症基因组图谱数据库对14个生物钟基因(Per1、Per2、Per3、Cry1、Cry2、Timeless、CLOCK、BMAL1、NPAS2、NR1D1、NR1D2、DEC1、DEC2和RO-RA)的RNA-Seq在6种腹部恶性肿瘤(肝癌、胃癌、胰腺癌、肾癌、直肠癌、结肠癌)组织及邻近正常组织中的表达进行分析,用Bioconductor的edgeR去标准化RNA-Seq数据.结果 14个生物钟基因在6种腹部恶性肿瘤组织中均低表达.与邻近正常组织比较,肝癌组织Cry2、CLOCK、Timeless、NPAS2、NR1D1、DEC1、DEC2、RORA基因低表达(P均<0.05),肾癌组织Per1、Per2、Cry1、Cry2、CLOCK、Timeless、NPAS2、NR1D1、NR1D2、DEC2、RORA基因低表达(P均<0.05),结肠癌组织Per1、Per2、Per3、Cry1、Cry2、Timeless、BMAL1、NR1D2、DEC1、DEC2、RORA基因低表达(P均<0.05),直肠癌组织Per1、Per3、Cry1、Cry2、CLOCK、BMAL1、NPAS2、NR1D1、DEC2、RORA基因低表达(P均<0.05),胃癌组织Per1、Per3、Cry2、CLOCK、Timeless、BMAL1、NPAS2基因低表达(P均<0.05).结论 生物钟基因在腹部恶性肿瘤组织中低表达,并且在不同类型腹部恶性肿瘤组织中表达亦存在一定差异.
