兔激素性股骨头缺血性坏死模型BMSCs超微结构观察

         

摘要

Objective To study the ultrastructure changes of bone mesenchymal stem cells (BMSCs) in steroid-induced avascular necrosis of the femoral head(SANFH) rabbit, so as to investigate the pathogenesis of SANFH. Methods The SANFH model of rabbit was established by using horse serum combining prednisolone acetate, Percoll was used to separate the bone mesenchymal stem cells in proximal femoral, and transmission electron microscopy was used to observe the ultrastructure. Results There was large number of lipid droplets in the proximal femoral of bone marrow, and chromatin condensation was found in BMSCs; large number of lipid droplets and mitochondria swelling were found in the cytoplasm.Conclusions The probable mechanism of steroid-induced SANFH maybe influence aerobic respiration of cells and lipid metabolism of BMSCs by disturbing hematological system.%目的 观察兔激素性股骨头缺血性坏死(SANFH)模型中骨髓间充质干细胞(BMSCs)的超微结构变化,探讨SANFH的发生机制.方法 采用马血清加醋酸泼尼松龙制备兔SANFH模型,采用Percoll细胞分离液分离其股骨近端BMSCs,透射电镜观察其超微结构.结果 骨髓内出现大量脂肪滴,其中BMSCs 出现胞核染色质浓缩;胞质中出现大量脂滴及线粒体肿胀.结论 激素导致SANFH的可能机制为通过对血液系统的干扰,影响股骨头内各种细胞的有氧呼吸;又通过干扰BMSCs 的脂类代谢影响骨的修复,最终导致股骨头坏死塌陷.
