木犀草素对人胃癌HGC-27细胞迁移的影响及机制

         

摘要

Objective To investigate the inhibition effect of luteolin on the migration of human gastric cancer cell line ( HGC-27 cells) in vitro and its mechanism. Methods The effect of the different concentration of luteolin on the experimental metastasis ability of HGC-27 cells was observed by scratch test and Tran swell metastasis experiments. The level of MMP-7 mRNA in HGC-27cell was detected by using Western Blot. Results The HGC-27 cell area on scratch was 96. 2% , 72. 3% and 55. 6% after treated with luteolin at 24 hours time later, while it was a totally coverage of HGC-27 cell without adopted by luteolin. The number of HGC cells passing through filter membrane after treated with luteolin decreased than those without. The levels of MMP-7 protein in HGC-27 cells decreased significantly after treated with the luteolin. Conclusion The luteolin inhibit the migration ability of HGC-27 cells in vitro by which inhibit the MMP-7 protein level of HGC-27 cell.%目的 观察木犀草素对人胃癌细胞系HGC-27细胞迁移能力的影响,并探讨其作用机制.方法 用10、20、40 μmol/L木犀草素培养HGC -27细胞.用划痕实验、Transwell实验观察不同浓度木犀草素对同样条件培养下胃癌HGC-27细胞迁移能力的影响,并采用Westem blot方法检测金属基质蛋白酶-7(MMP-7)mRNA.结果 划痕实验中培养24h后,未经木犀草素处理的HGC-27细胞运动迁移基本完全覆盖了划痕区域,而经10、20、40 μmol/L 木犀草素处理的HGC-27细胞仅覆盖了96.2%、72.3%、55.6%的划痕区域.Transwell实验中经10、20、40 μmol/L木犀草素处理的HGC细胞穿膜数量占对照组细胞穿膜数量的百分比分别为97.6%、59.5%、20.0%.经10、20、40μmol/L木犀草素处理的HGC-27细胞内MMP-7蛋白的表达水平明显降低,并与木犀草素的浓度呈负相关.结论 木犀草素抑制了胃癌HGC-27细胞的迁移能力,其作用与抑制HGC-27细胞内的MMP-7蛋白表达水平有关.
