目的 探讨人参皂苷Rg1联合骨髓间充质干细胞(BMSCs)对大鼠脑缺血再灌注损伤的保护作用.方法 25只大鼠随机被分为正常对照组(A组)、模型组(B组)、BMSCs治疗组(C组)、人参皂苷Rg1治疗组(D组)、BMSCs和人参皂苷Rg1联合治疗组(E组)各5只.采用大脑中动脉栓塞法制备脑缺血再灌注模型,利用BrdU标记BMSCs后行脑室内移植.于造模后4周采用单盲法进行神经功能缺损评分后,用焦油紫染色检测大脑皮层神经元,用免疫组化染色检测BrdU、NSE的表达情况.结果 A组未见神经功能缺损症状,皮层神经元细胞质中可见深紫色尼氏体；B组呈明显的神经功能缺损症状,尼氏体明显减少甚至消失；与B组比较,C、D、E组神经功能缺损症状改善、神经元存活数量增高(P均＜0.05),其中E组最为显著；免疫组化染色C、E组可见梗死区周围有BrdU阳性细胞；A组NSE阳性表达,B组少量表达,与B组比较,C、D、E组NSE阳性表达增强、以E组最为显著.结论 人参皂苷Rg1联合BMSCs能够促进脑缺血再灌注损伤的修复.%Objective To investigate the protective effect of ginsenoside Rgl jointed BMSCs on ischemia and reperfusion injury in rats. Methods A total of 25 SD rats were divided into normal control group (group A) , model group ( group B) , BMSCs treatment group ( group C) , ginsenoside Rgl treatment group ( group D) , BMSCs and ginsenoside Rgl combination therapy group ( group E) , each group 5 rats. Model of cerebral ischemia and reperfusion was established by the method of middle cerebral artery occlusion. Intraventricular transplantation was done after BrdU labeling BMSCs. After 4W, single-blinded neurological deficit scores was detected. The expressions of BrdU and NSE protein were detected by Nissl staining counted cortical neurons and immunohistochemical staining. Results There was no neurological deficit symptoms in Group A . Deep purple Nissl bodies can be seen in the cortical neurons of the cytoplasm. It showed a significant neurological deficit symptoms in group B. Nissl bodies were significantly reduced or even disappear. Compared with group B, the neuronal number in the group C, D, E increased, especially in group E. BrdU positive cells can be seen a-round the infarcted area in group C and E. Expressions of NSE in group C, D and E were higher than those in group B, especially in group E. Conclusion Ginsenoside Rgl jointed BMSCs can promote the repair of cerebral ischemia and reperfusion injury.
