红芪总多糖对衰老大鼠体内自由基清除及抗氧化能力的调节作用

     

摘要

Objective To observe the regulation function of total hedysari polysaccharide ( THPS) in sweeping away free radicals and antioxidant capacity on aging rats .Methods Seventy-two Wistar rats were randomly divided into groups A, B, C, D, E and F.Every group had 12 rats.The aging models were made in groups A , B, C, D and E.and group F was the control group .On the next day of modeling , rats in the groups A , B, C and D were intraperitoneally injected with THPS 50, 100, 200 and 400 mg/kg every day respectively for 30 days.Rats in groups E and F were intraperitoneally injec-ted with 10 mL/kg normal saline everyday for 30 days.The scavenging rate of superoxide anion free radical (O·-2 ) and the hydroxyl free radical ( OH-· ) in each group was detected .The content of malondialdehyde ( MDA) , the activities of su-peroxide dismutase (SOD) and glutathione peroxidase (GSH-PX) in the blood serum and hepatic tissues were detected . Results Compared with group E , the scavenging rates of O ·-2 and OH-· were higher in groups A , B, C and D ( all P<0.05);the content of MDA in the blood serum was decreased and the activity of GSH-PX in the blood serum was higher in the group B (all P<0.05);in the blood serum and hepatic tissues, the content of MDA was all decreased and the activi-ties of SOD and GSH-PX were increased in groups C and D (all P<0.05).Conclusion THPS can sweep away the con-tent of O ·-2 and OH -· , decreases MDA in the blood serum and hepatic tissue homogenate , and increases the activities of SOD and GSH-PX in the blood serum and hepatic tissue homogenate .%目的:观察红芪总多糖(THPS)对衰老大鼠体内自由基清除及抗氧化能力的调节作用。方法72只Wistar大鼠随机分为A、B、C、D、E、F组各12只,A、B、C、D、E组均制备衰老模型,F组不制备模型;造模后第2天, A、B、C、D组大鼠分别经腹腔注射50、100、200、400 mg/kg THPS,E、F组大鼠腹腔注射10 mL/kg生理盐水,连续30 d。比较各组大鼠体内超氧阴离子(O·-2)、羟自由基(OH-·)清除率,检测血清和肝组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-PX)活性。结果与E组比较,A、B、C、D组体内O·-2、OH-·清除率高(P均<0.05);与E组比较,B组血清MDA含量降低,血清GSH-PX活性增强(P均<0.05);与E组比较,C、D组血清和肝组织MDA含量降低,血清和肝组织SOD活性、GSH-PX活性提高(P均<0.05)。结论THPS可清除大鼠体内O·-2、OH-·,降低肝组织和血清中MDA含量,提高SOD、GSH-PX活性。
