高效液相色谱法分离和测定油茶茎尖组织中4种内源激素

         

摘要

A reversed - phase high performance liquid chromatography ( RP - HPLC ) method was developed for the determination of indole - 3 - acetic acid (IAA), gibberellin acid (GA3) , zeatin riboside (ZR), and abscisic acid ( ABA) in Camellia oleifera shoot - tip tissues. The method was performed with a reverse -phase C18 column and using the mixture of methanol and water ( containing 0.75% acetic acid) with the volume ratio of 35: 65 as mobile phase; the sample injection volume was 10 μl and the flow rate was 0.8 ml/min; the detection wavelength was 254 nm and the column temperature was 30℃. These hormones were separated well by this method. The lowest detection limit of IAA, GA3, ZR and ABA were 0. 05, 0. 01, 0. 02, and 0.01 mg/L respectively and the recovery rate were 98.2% , 97.9% , 96. 3% and 93.4% respectively. In a word, the method was convenient and accurate.%建立了高效液相色谱法测定油茶茎尖组织中生长素(IAA)、赤霉素(GA3)、玉米核苷(ZR)、脱落酸(ABA)4种内源激素的方法.应用反相C18色谱柱,以甲醇-水(含0.75%冰乙酸,35∶65,V/V)为流动相,流速为0.8 ml/min,进样量10μl,检测波长254 nm,柱温30℃,该方法各峰分离效果理想.IAA、GA3、ZR、ABA的最低检出限分别为0.05、0.01、0.02、0.01 mg/L,回收率分别为98.2%、97.9%、96.3%、93.4%.该方法易操作且准确性高.
