Objective:To investigate the effect of evodiamine (Evo) on the expresstion of peptide NPY in the arcuate nucleus (ARC) of the hypothalamus in rats .Methods:Male Sprague‐Dawley rats (5 weeks old ,n=36) were randomly divided into three groups (n=12) .Intragastric administration of Evo 40 mg/kg ,Evo 4 mg/kg or water was infused for 25 days ,once a day .We evaluated the effect of Evo on NPY peptide expression .NPY was de‐tected by immunohistochemistry in cytoplasm of the hypothalamic ARC .The expression of peptide NPY in hypotha‐lamic ARC was measured by Western Blotting .Results :The frequency of NPY‐IR neurons of Evo 40 mg/kg was significantly smaller than control group .The expression of orexigenic peptide NPY in hypothalamic ARC of the ani‐mals treated with Evo 40 mg/kg (Ratio of light density between NPY and GAPDH) had a noticeable drop compared with the control group .Conclusion:Our results show that intragastric administration of Evo (40 mg/kg) decreased orexigenic NPY peptide level in the arcuate nucleus (ARC) of the hypothalamus .This is one of the most important mechanisms of Evo in the hypothalamus .%目的:研究吴茱萸碱(Evo)对大鼠下丘脑弓状核神经肽Y(NPY)蛋白表达水平的影响。方法:选用雄性5周龄Sprague‐Dawly大鼠36只,分为对照组、Evo 4mg/kg组、40 mg/kg组。Evo灌胃25d后取材;采用免疫组织化学和蛋白免疫印迹的方法,观察 Evo 对下丘脑弓状核中NPY免疫阳性产物的表达和NPY蛋白水平的影响。结果:吴茱萸碱40 mg/kg干预后,下丘脑弓状核NPY‐IR阳性细胞数显著减少,同时NPY蛋白表达水平显著降低。结论:Evo可显著降低大鼠下丘脑弓状核促食欲肽N PY蛋白水平,提示下丘脑可能是Evo调节摄食行为的重要中枢。
