蒺藜总皂苷对大鼠肺缺血再灌注损伤后细胞凋亡的影响

摘要

Objective:To investigate the effects of gross saponin from tribulus terrestris (GSTT ) on pneumocyte apoptosis in lung ischemia‐reperfusion injury .Methods:Thirty healthy rats were randomly divided into three groups:C group ,IR group ,G group .After 120 min reperfusion ,TUNEL assay was applied to detect lung apop‐totic index(AI) and immunohi stochemical SP technique was used to detect the expressions of Bcl‐2 and Bax protein . Results:GSTT could decrease AI(P<0 .05) ,and lower Bax protein expression(P<0 .05) .Conclusion:GSTT can re‐duce lung apoptosis through regulating protein expressions of Bcl‐2 and Bax ,so that it provides significant protective effects on lung ischemia‐repe rfusion injury .%目的：探讨蒺藜总皂苷（GSTT）对大鼠肺缺血再灌注损伤后细胞凋亡的影响。方法：建立在体肺缺血再灌注模型。将30只大鼠随机分为正常对照组（C组），缺血再灌注组（IR组），蒺藜总皂苷组（G组）。每组分别在再灌120min时处死10只大鼠，留取右肺组织，原位细胞凋亡检测（TUNEL）法检测肺组织中细胞凋亡指数（AI）及免疫组化法检测肺组织Bcl‐2、Bax 蛋白的表达。结果：IR组肺组织AI、Bax／Bcl‐2的表达均明显升高，而预先给予GST T 干预后，G组均可以减弱缺血再灌注诱导上述指标的变化。结论：GS T T可通过下调Bax／Bcl‐2蛋白表达，从而抑制细胞凋亡，对肺缺血再灌注损伤起到保护作用。
