大鼠脑梗死模型神经行为学评价的可靠性探讨

             

摘要

目的:探讨大鼠大脑中动脉阻塞模型(MCAO)神经行为学评价的可靠性。方法:制备大鼠MCAO模型,清醒后即刻(D0)行神经行为学评分及MRI成像,术后24h(D1)再次行神经行为学评分,后断头取脑行T TC染色。结果:手术组12只大鼠均有程度不等的神经缺损症状,DWI显示大面积或局限性的缺血损伤区域,10只大鼠 TTC染色可见程度不等的梗死病灶。相关分析提示D0时间点Longa评分、Garcia评分与根据磁共振弥散加权成像ADC图计算的缺血梗死体积均无显著相关。D1时间点Longa评分、Garcia评分与根据 T TC染色计算的脑梗死体积均呈显著相关。结论:在大鼠MCAO模型超急性期神经行为学评分可靠性较差,术后24h神经行为学评分与脑梗死严重程度相关性良好。%Objective:To assess the reliability of behavioral testing in experimental rats model of ischemia stroke .Methods :Sprague‐Dawley rats were subjected to 90 min of MCAO ,then were evaluated with behavioral tes‐ting and scanned with MRI after they woke up .The behavioral testing were repeated 24 h after operation ,followed by sacrifice .The cerebral infarction were detected by TTC examination .Results:In operated group ,twenty rats have neural function deficient in varying degrees ,and demonstrated massive or focal high signal on DWI ,ten of them showed red cerebral infarction lesions on TTC staining .The scores of behavioral testing were correlated with infarc‐tion size on TTC staining on D1 ,conversely ,No correlation was found between the scores on D0 and infarction size on ADC map .Conclusion:The behavioral testing fails to evaluate the neurologic deficient in super acute period of cerebral infarction of rats .There are significant correlation between behavioral score and cerebral infarction size at 24 h after operation .
