重组人生长激素对心力衰竭大鼠心脏结构及功能的影响

             

摘要

目的:探讨重组人生长激素(rh‐GH)对心力衰竭大鼠心脏结构及功能的影响。方法:将30只清洁级 SD 大鼠分为3组,即空白组、模型组与 rh‐GH 组,各10只。空白组不予处理,常规喂养;模型组、rh‐GH 组均采用阿霉素法建立心力衰竭大鼠模型;模型组建模后每日皮下注射等剂量生理盐水;rh‐GH 组建模后每日皮下注射0.4 U /kg rh‐GH ,均连续注射1个月。采用心脏彩色多普勒诊断仪测定各组大鼠心脏功能的变化,记录 LVDD 、LVSD 、LVPWT 、IVS 、EF ,测定 LVSP 、CVP 、LVEDP ,取颈动脉血测定 NT‐proBNP 、TNF‐α、cTn Ⅰ水平,并处死大鼠,取心脏组织,观察心肌超微结构的变化,测定心肌间质纤维化程度。结果:与空白组比较,模型组、rh‐GH 组 LVDD 、LVSD 增加,LVPWT 、IVS 、EF 降低( P <0.05);与模型组比较,rh‐GH 组 LVDD 、LVSD 降低、LVPWT 、IVS 、EF 上升(P<0.05)。与空白组比较,模型组、rh‐GH 组 LVSP 降低,CVP 、LVEDP上升(P<0.05);与模型组比较,rh‐GH 组 LVSP 上升,CVP 、LVEDP 降低(P <0.05)。与空白组比较,模型组、rh‐GH 组 LVSP 降低,cTn Ⅰ、NT‐pro BNP 、TNF‐α均上升(P <0.05);与模型组比较,rh‐GH 组 cTn Ⅰ、NT‐pro BNP 、TNF‐α均降低(P<0.05)。与空白组比较,模型组、rh‐GH 组心肌间质纤维指数上升(P<0.05);与模型组比较,rh‐GH 组心肌间质纤维指数下降(P <0.05)。结论:rh‐GH 可改善心力衰竭大鼠心脏功能、心脏结构,减轻心肌间质纤维化程度。%Objective :To investigate the effect of recombinant human growth hormone (rh‐GH ) on cardiac structure and function in rats with heart failure .Methods :Thirty clean SD rats were divided into 3 groups ,namely , the blank group ,the model group and the rh‐GH group ,10 rats in each group .The blank group was not treated and was given conventional feeding ;The model group and rh‐GH group were used to establish rat model of heart failure with adriamycin .After modeling ,the model was subcutaneously injected with isodose normal saline every day while rh‐GH group was subcutaneously injected with 0 .4 U /kg rh‐GH and were injected for 1 month .The color Doppler echocardiography was performed to determine the changes of cardiac function in rats .LVDD ,LVSD ,LVPWT ,IVS and EF were recorded .LVSP ,CVP and LVEDP were determined .The carotid arterial blood was collected to deter‐mine the levels of NT‐proBNP ,TNF‐α and cTnI .Then ,the rats were killed and heart tissues were taken to observe the changes of myocardial ultrastructure and determine the degree of myocardial interstitial fibrosis .Results :Com‐pared with the blank group ,LVDD and LVSD of the model group and rh‐GH group were higher while LVPWT , IVS and EF were lower (P< 0 .05) .Compared with model group ,LVDD and LVSD of rh‐GH group were lower while LVPWT ,IVS and EF were higher (P< 0 .05) ;Compared with the blank group ,LVSP of the model group and rh‐GH group were lower while CVP and LVEDP were higher (P< 0 .05) .Compared with model group ,LVSP of rh‐GH group was higher while CVP and LVEDP was lower (P< 0 .05) ;Compared with the blank group ,LVSP of the model group and rh‐GH group were lower while cTn I ,NT‐proBNP and TNF‐α were higher ( P < 0 .05) . Compared with the model group ,cTn I ,NT‐proBNP and TNF‐α in rh‐GH group were lower (P< 0 .05) ;Compared with the blank group ,myocardial interstitial fibrosis indexes of the model group and rh‐GH group increased (P <0 .05) .Compared with the model group ,interstitial fibrosis indexes of the rh‐GH group decreased (P < 0 .05) . Conclusion :Rh‐GH can improve cardiac function and structure in rats with heart failure and reduce the degree of my ‐ocardial interstitial fibrosis .
