PEG和钙对栀子花粉萌发的影响

         

摘要

以栀子(Gardenia jasminoides Ellis)花粉为试验材料,采用花粉人工培养法研究了培养基中PEG和钙对栀子花粉萌发的影响.结果表明,不同分子量的PEG对栀子花粉离体萌发率的影响不同,PEG-6000较其他3种分子量的PEG促进花粉萌发的效果要好;PEG4种分子量的最适浓度不同,其中PEG-4000和PEG-6000促进栀子花粉萌发的最佳浓度为15g/L,而PEG-2000和PEG-10000促进栀子花粉萌发的最佳浓度分别为20g/L和10g/L.以0.8%的琼脂+30mg/L蔗糖+15mg/L硼酸+15g/L PEG-6000为基本培养基组分,分别添加不同浓度的CaCl2,发现适宜浓度的钙有利于栀子花粉萌发,但钙浓度过高抑制萌发,栀子花粉萌发的最适Ca2+浓度为2.0mmol/L.%Cape jasmine ( Gardenia jasminoides Elli) was used to study the effects of PEG and calcium on pollen germination characteristics of Gardenia jasminoides Elli with the method of pollen artificial culture. The results showed that the optimal concentrations of different PEG were different and the optimal concentration of PEG-4000 and PEG-6000 to promote pollen germination were 15 g/L,the optimal concentration of PEG-2000 and PEG-10000 to promote pollen germination were 10 g/L. 0. 8% agar +30 mg/L sucrose + 15 mg/L boric acid +15 g/L PEG-6000 as the basic medium components, different concentration of CaCl2 was added to the medium, the results showed that the suitable concentration of calcium promoted pollen germination, but the high concentration of calcium inhibited the pollen germination. The optimal concentration of Ca + for Cape jasmine pollen germination was 2.0 mmol/L.
