阳和汤对兔膝骨关节炎模型关节液中SOD、NO的影响

         

摘要

Objective:To study the mechanism of Yanghe Decoction(阳和汤) in the prevention and treatment of osteoarthritis by observing its effect on SOD and NO in the synovial lfuid of rabbits with knee osteoarthritis.Methods:24 healthy New Zealand rabbits were randomly divided into a Yanghe Decoction group,a positive control group,a model control group and a blank control group,6 rats in each.Except for the blank control group,the other groups were used to establish ostarthritis models with papain.The Yanghe Decoction group was given intragastric administration with Yanghe Decoction,the positive control group with Jingu Tongxiao Pill (筋骨痛消丸) and the model control group and blank control group with physiological saline,once a day for ifve weeks.HE staining was used to observe the changes of the morphosis of cartilage,and ELISA was used to detect the changes of SOD and NO in synovial lfuid.Results:HE staining:The cartilage morphosis in the Yanghe Decoction group and the positive control group was significantly better than that of the model control group. Mankin score of articular cartilage tissue injury:Compared with the blank control group,the integrals in the other three groups increased signiifcantly(P< 0.05);the differences of that between Yanghe Decoction group,the positive control group and the model control group were statistically signiifcant(P< 0.05);the difference of that between the Yanghe Decoction group and the positive control group was not statistically signiifcant(P> 0.05).ELISA:The content of SOD in the Yanghe Decoction group and the positive control group was higher than that in the model control group,the difference being statistically signiifcant(P< 0.05);the content of NO in the Yanghe Decoction group and the positive control group was lower than that in the model control group,the difference being statistically signiifcant(P< 0.05).Conclusion:Yanghe Decoction can be used to treat knee osteoarthritis by increasing the content of SOD and decreasing the content of NO in the synovial lfuid.%目的:通过观察阳和汤对兔膝骨关节炎模型关节液中超氧化物歧化酶(SOD)、一氧化氮(NO)的影响,探讨阳和汤防治骨关节炎的作用机制。方法:将24只健康新西兰兔随机分为阳和汤组、阳性对照组、模型对照组、空白对照组,每组6只。除空白对照组外,其余各组均采用木瓜蛋白酶造模方法建立骨关节炎模型。阳和汤组以阳和汤灌胃,阳性对照组以筋骨痛消丸灌胃,空白对照组、模型对照组以生理盐水灌胃,每日1次,持续给药5周。采用HE染色观察软骨形态结构的变化,用酶联免疫吸附法(ELISA法)检测关节液中SOD、NO含量的变化。结果:HE染色:软骨形态结构阳和汤组与阳性对照组明显优于模型对照组。关节软骨组织损伤Mankin's评分:与空白对照组比较,其余3组评分明显增加(P<0.05);阳和汤组、阳性对照组与模型对照组比较,差异有统计学意义(P<0.05);阳和汤组与阳性对照组比较,差异无统计学意义(P>0.05)。ELISA检测法:阳和汤组与阳性对照组关节液中SOD含量均高于模型对照组,差异有统计学意义(P<0.05);阳和汤组与阳性对照组关节液中NO含量均低于模型对照组,差异有统计学意义(P<0.05)。结论:阳和汤可以通过升高兔膝骨关节炎模型关节液中SOD含量,降低其NO含量,从而对膝骨关节炎起到治疗作用。
