薄膜超声法制备龙胆苦苷脂质体

摘要

Objective To prepare the liposomes of Gentiopicrin by the film-ultrasonic dispersion method. Methods Preparing the liposomes of Gentiopicrin by the film-ultrasonic dispersion method, using HPLC to determine the content of Gentiopicrin, and mini-column centrifugation was used to separate liposomes to determine the envelopment rate. Results The proportion of the cholesterol to phospholipid was 1:2,the pH value of buffer solution was 5.7, the proportion of the octadecyl amine to phospholipid was l:10,the proportion of the drug to phospholipid was 1:12. The envelopment rate of the liposomes of Gentiopicrin prepared by the optimum process reached 75.14%.Conclusion The established method could prepare the liposomes of Gentiopicrin with good envelopment rate.%目的制备均匀稳定的龙胆苦苷脂质体.方法 采用薄膜超声法制备龙胆苦苷脂质体；采用HPLC法测定含量,色谱条件:流动相为甲醇:水=30∶70,流速为1 mL/min,检测波长为270 nm:采用微柱离心法分离脂质体测量包封率.结果 胆固醇:磷脂比1∶2,PBS缓冲液的pH值为5 7,十八胺磷脂比1∶10,药脂比为1∶12.最优制备工艺采用薄膜超声法制备龙胆苦苷脂质体最佳包封率为75.14％.结论 采用薄膜超声法制备龙胆苦苷脂质体包封率较好.
