目的 探讨澳洲茄边碱抑制CNE-1细胞转移的分子机制.方法 CCK8活力检测法和transwell实验分别观察澳洲茄边碱对CNE-1细胞增殖、侵袭和迁移的影响;Real-time quantitative PCR和蛋白质印迹法探索澳洲茄边碱对snail、slug、E-cadherin和N-cadherin m RNA及蛋白表达的影响.结果澳洲茄边碱以浓度和时间依赖性抑制CNE-1细胞增殖 (P<0.05);以浓度依赖性对CNE-1细胞的侵袭和迁移能力进行抑制 (P<0.05);分别在基因和蛋白质水平上抑制snail、slug、E-cadherin和N-cadherin的表达水平 (P<0.05).结论 澳洲茄边碱通过在基因和蛋白质水平抑制转录因子snail、slug的表达, 进而影响E-cadherin和N-cadherin的表达, 从而抑制CNE-1细胞的转移.%Objective To discuss the molecular mechanism of Solamargine (SM) in inhibiting CNE-1 metastasis. Methods The inhibition of SM in CNE-1 proliferation, invasion and migration are discussed by CCK8 assay and transwell assay respectively. The effects of SM on snail, slug, E-cadherin and N-cadherin m RNA and protein expression are tested by Real-time quantitative PCR and western blot assay. Results The CNE-1 cells proliferation is inhibited by SM depended on concentration and time (P < 0.05). The CNE-1 cells invasion and migration are inhibited by SM depended on concentration (P < 0.05). The m RNA and protein expression of snail, slug, E-cadherin and N-cadherin are inhibited by SM (P < 0.05). Conclusion The inhibition of CNE-1 metastasis by SM is through inhibiting the expression of snail and slug, and mediated the downstream proteins' expression of E-cadherin and N-cadherin.
