苎麻胚珠的酶解法提取及其整体染色透明技术

摘要

In order to observe the embryonic growth in Boehmeria tricuspids better, the ovules were isolated from the female flowers by enzymatic method and the corresponding whole stain - clearing technique was studied. Results showed that a mixture solution of 3% cellulase and 1% pectinase had the best dissociation effect, and staining with hematoxylin, clearing with methyl salicylate were suitable for observing the ovules.%本文以悬铃叶苎麻雌花为试验材料,研究通过酶解方法分离出胚珠并对其进行整体染色透明的技术,目的在于提高整体透明技术在苎麻胚胎发育观察中的效果.结果表明:3％纤维素酶与1％果胶酶混合酶液提取胚珠的效果最好,采用苏木精染色后冬青油透明的方法观察效果较好.
