毛细管电泳法测定中药篇蓄中山奈素、槲皮素和咖啡酸

     

摘要

A method for the simultaneous separation and determination of kaempferol, quercetin and caffeic acid in the TCM, Polygonum aviculare by capillary electrophoresis was proposed. Mixed solution of 20 mmol·L^-1 Na2 B4 O7-NaH2 PO4 of pH 8.5 was used as the buffer solution, moving voltage of 20 kV and gravity injection time of 14 s were applied. UV-deteetion at the wavelength of 254 nm was adopted. The 3 analytes were completely separated within 7 min. Linearity ranges between 0. 50-100 (for kaempferol), 0. 70-146 (for quereetin) and 1.0-124 mg·L^-1 (for eaffeic acid) were obtained, with detection limits (3S/N) of 0. 3, 0. 4 and 0. 6 mg·L^-1 respectively. The proposed method was applied to the analysis of sample of Polygonum aviculare, giving values of recovery obtained by standard addition method in the range of 95.3%-106% with RSD's (n= 5) less than 3. 5 %.%提出了毛细管电泳法同时分离测定中药篇蓄中山柰素、槲皮素和咖啡酸含量的方法,以pH8.5的20mmol·L^-1硼酸钠-磷酸二氢钠为缓冲溶液,运行电压20kV,重力进样时间14S,在254nm波长处进行紫外检测。山柰素、槲皮素和咖啡酸在7min内即可实现分离。3者的线性范围依次为0.50~100,0.70~146,1.0~124mg·L^-1,检出限(3S/N)依次为0.3,0.4,0.6mg·L^-1。采用此法对篇蓄样品进行了分析,所得加标回收率在95.3%~106%之间,测定值的相对标准偏差(n=5)均小于3.5%。
