蒸发光散射测定扶正益胃口服液中黄芪甲苷的含量

摘要

目的 应用高效液相色谱蒸发光散射(HPLC-ELSD)检测法建立扶正益胃口服液中黄芪甲苷的含量测定方法.方法 采用Extend-C18(250 mm×4.6 mm,5μm)色谱柱,流动相:乙腈-水(35∶65),流速:1.0 ml ·min-1,蒸发光散射检测器(漂移管温度:50℃,增益:6),柱温:25℃.结果 黄芪甲苷在0.05 ～1.00 mg ·ml-1范围内,其常用对数与峰面积常用对数存在良好的线性关系,r =0.9997,样品平均回收率为100.4％(RSD=0.9％,n=9).结论 建立了HPLC-ELSD法测定扶正益胃口服液中黄芪甲苷含量的方法,且方法简单、方便、专属性高,适合该制剂中黄芪含量测定.%Objective To establish a method for determining the content of astragaloside Ⅳ in Fuzheng Yiwei oral liquid by HPLC-ELSD.Methods Extend-C18 (250 mm × 4.6 mm,5 μm) was used as the chromatography column.The mobile phase consisted of acetonitrile-water (35∶ 65) at a flow rate of 1.0 ml ·min-1.Column temperature was 25 ℃.Results Astragaloside Ⅳ showed a good linear relationship in the range of 0.05-1.00 mg ·ml-1,r =0.9997.The average recovery was 100.4％ (RSD =0.9％,n =9).Conclusion This method can accurately detect the content of astragaloside in Fuzheng Yiwei oral liquid.
