一测多评法测定藏药五味黄连丸中4种生物碱的含量

摘要

目的 建立藏药五味黄连丸(黄连、红花、诃子、渣驯膏和麝香)中表小檗碱、黄连碱、巴马汀和小檗碱的一测多评含量测定方法.方法 采用RP-HPLC法,以乙腈-水(1 mL·L-1三乙胺和1 mL·L-1甲酸)(30∶70)为流动相;检测波长:345 nm;柱温:25 ℃;流速:0.8 mL·min-1;以盐酸小檗碱为内标物,测定小檗碱、表小檗碱、黄连碱和巴马汀的相对校正因子,计算五味黄连丸中4种生物碱的含量,并比较一测多评法的计算值与常规外标法的实测值的相似度.结果 一测多评法与常规外标法测得的结果差异无统计学意义,2组数据的夹角余弦值、相关系数和相似度均达到0.999.结论 利用一测多评法可简便可靠、快速、准确地控制五味黄连丸的质量.%Objective To estahlish a method for the simultaneous determination of epiberberine,coptisine,palmatine and berberine in Tibetan medicine of Wuweihuanglian Pills (Coptidis rhizoma,Crarthamus tinctorius,myrobalan,trogopterus dung cream and musk) by quantitatively analyzing multi-components with a single-marker (QAMS).Methods RP-HPLC method was adopted.The mobile phase was acetonitrile-water (1 mL·L-1 triethylamine and 1 mL·L-1 formic acid) (30∶70),the flow rate was 0.8 mL·min-1with UV detection wavelength at 345 nm and the column temperature was maintained at 25 ℃.With berberine hydrochloride as an internal standard,the relative correlation factors (RCF) of epiberberine,coptisine and palmatine to berberine were determined.The content of these 4 alkaloids were determined by external standard method and QAMS,respectively.The similarity of those data was compared.Results No significant difference in assay results was observed.The included angle cosine, correlation and similarity all reached 0.999.Conclusion It is simple,reliable,rapid and accurate to evaluate the quality of Wuweihuanglian Pills by QAMS.
