神经导管联合人雪旺细胞修复大鼠外周神经损伤的研究

     

摘要

目的:探讨神经导管联合人雪旺细胞修复大鼠外周神经损伤的作用。方法:60只大鼠建立外周神经损伤模型,随机分为自体神经移植组、单纯导管修复组、联合修复组各20只,分别采取自体神经移植、单纯导管修复、雪旺细胞联合神经导管修复治疗,治疗后13周测定坐骨神经功能指数(SFI)、Jitter 值,甲苯胺蓝染色观察再生神经中段髓鞘化轴突。结果:治疗后第13周,自体神经移植组的 SFI 和 Jitter 值小于联合修复组,联合修复组的 SFI 和 Jitter 值小于单纯导管修复组,差异均有统计学意义(P<0.05);修复后,自体神经移植组、单纯导管修复组、联合修复组的髓鞘厚度分别为(3.01±0.27)、(2.22±1.10)、(2.63±1.17)μm,联合修复组的髓鞘厚度小于自体神经移植组,大于单纯导管修复组,差异有统计学意义(P<0.05)。结论:神经导管联合人雪旺细胞修复大鼠外周神经损伤有效。%Objective:To investigate the effect of nerve conduits combined with human Schwann cells on repair of peripheral nerve injury in rats. Methods: The peripheral nerve injury models were established in 60 rats, which were randomly divided into autologous nerve graft group (given autologous nerve graft), pure conduit repairment group (given pure conduit repairment), and combined remediation group (given Schwann cells combined with conduit re-pairment), with 20 rats in each group. Sciatic Functional Index (SFI) and Jitter values were examined after 13 weeks of treatment, and the axonal regeneration was observed by toluidine blue staining. Results: After 13 weeks of the treatment, the SFI and Jitter values in the autologous nerve graft group were significantly lower than those in the combined remediation group, while the SFI and Jitter values in the combined remediation group were significant lower than those in the pure conduit repairment group (P<0.05). After repair, the myelin sheath thickness of the groups autologous nerve graft group, pure conduit repairment, combined remediation were (3.01±0.27), (2.22± 1.10), (2.63±1.17)μm respectively.The myelin sheath thickness of the combined remediation group was smaller than that of the autologous nerve graft group but greater than the pure conduit repairment group (P<0.05). Conclu-sion: The nerve conduit combined with human blood cells is safe and effective for repairing peripheral nerve injury.
