鄂尔多斯高原钝顶节旋藻SOD特性研究

         

摘要

In this study,the pyrogallol auto-oxidation method was used to investigate the characteristics of superoxide dismutase (SOD) of Arthrospira platensis from the Erdos Plateau.The maximum absorption wavelength for SOD was 320 nm in the ultraviolet range.The optimal pH in a Tris-HCl buffer and the optimal temperature for SOD were 8.2 and 25 ℃,respectively.The SOD activity was higher in a pH range of 6-10.Superoxide dismutase activity decreased at temperatures higher than 40 ℃ for more than 20 minutes and higher than 35 ℃ for more than 60 minutes.Similarly,SOD activity decreased at room temperature in light after a period of more than 2 days,almost not at all after seven days at room temperature in light,and exhibited absolutely no change at room temperature without light after nine days.%采用邻苯三酚自氧化法确定了鄂尔多斯高原钝顶节旋藻SOD特性.该SOD在紫外光区320 nm处有最大吸收值;在Tris-HCl缓冲溶液中最适pH值为8.2,最适温度为25℃;pH值稳定性范围为6~10;高于40℃的条件下保温20 min酶活性开始下降,高于35℃条件下保温60 min酶活性开始下降;室温下存放2d后活性开始下降,7d时活性基本丧失;室温下避光保存9d时SOD活性完全丧失.
