星点设计-响应面法优化黄柏提取工艺研究

摘要

目的：利用星点设计-响应面法优化黄柏的最佳提取工艺。方法：以盐酸小檗碱含量为评价指标，选取提取时间、液料比、提取次数3个因素进行 Box-Behnken 中心组合设计，通过星点设计-响应面法确定黄柏最佳提取工艺。结果：黄柏的最佳提取工艺为水煎煮提取，液料比8∶1，提取时间90 min，提取3次，盐酸小檗碱得率为32.15 mg·g －1。结论：采用星点设计-响应面法优化黄柏提取工艺，方法简单合理，且具有良好的预测性。%Objective:To optimize extraction process of Cortex Phellodendri Chinensis by central composite design-response surface methodology.Methods:Water was chose as the extraction solvent and the effect of three factors such as solvent-solid ratio,extraction time and times was designed by Box-Behnken central composite,taking the content of berberine hydrochloride as evaluation indicator.Extraction process was optimized by central composite design-response surface methodology.Results:The optimizing extraction process of Cortex Phellodendri Chinensis were as follows:ratio of liquid to solid was 8:1,extraction repeated 3 times,90 minutes every time.The yield of berberine hydrochloride under this condition was 32.15 mg·g -1.Conclusion:Optimizing extraction process of Cortex Phellodendri Chinensis by central composite design-response surface methodology is reasonable,simple,and has good predictability.
