Optimization for ultrahigh pressure extraction of berberine from Cortex Phellodendri by central composite design-response surface methodology

As a novel extraction technique, ultrahigh pressure extraction (UPE) was used to extract berberine from?Cortex Phellodendri?firstly, which procedure has higher extraction yield, lower extracting temperature, shorter extracting time and less power consumption. The content of berberine was determined by reverse phase-high performance liquid chromatography (RP-HPLC). The UPE process of berberine from?Cortex Phellodendri was optimized by using central composite design-response surface methodology (CCD-RSM). The results showed that the optimal conditions for the optimum extraction conditions were ethanol concentration of 69.1%, liquid-solid ratio of 31.3, extracting pressure of 243.30 MPa, extraction time of 2 min, the extraction efficiency of berberine was 7.70 mg·g-1, which was higher than conventional extractions. We found that UPE had provided a novel method for extraction of berberine from?Cortex Phellodendri.
