RP-HPLC法同时测定枫蓼肠胃康胶囊中4种黄酮苷成分的含量

摘要

目的:建立反相高效液相色谱法同时测定枫蓼肠胃康胶囊中芦丁、金丝桃苷、槲皮苷、山奈酚-3-O-芸香糖苷含量的方法.方法:采用Waters X bridge C1s(250 mm×4.6 mm,5μm)色谱柱;流动相系统:乙腈(A)-0.2％甲酸水溶液(B),梯度洗脱;流速:1.0 mL ·min-1;柱温:25℃;检测波长:350 nm.结果:芦丁、金丝桃苷、槲皮苷、山奈酚-3-O-芸香糖苷的质量浓度分别在12.42 ～310.50、5.95～ 148.75、6.85～171.25、7.88 ～197.00 μg ·mL与色谱峰面积呈良好的线性关系,加样回收率分别为98.6％、104.2％、104.5％、101.0％ (n =6),RSD分别为1.3％、1.1％、1.3％、1.6％.结论:本方法符合方法学验证要求,简便、快捷,可用于枫蓼肠胃康胶囊中4种黄酮苷成分的同时测定.%Objective:To establish an RP-HPLC method for simultaneous determination of rutin,hyperoside,quercetin-3-rhamnoside and kaempferol-3-O-rutinoside in Fengliao Changweikang Jiaonang.Methods:The Waters X bridge C18 (250 mm ×4.6 mm,5 μm)column was adopted;the mobile phase was methanol (A)-0.2％ formic acid (B) with gradient elution,flow rate was 1.0 mL · min-1,column temperature was 25 ℃,and detection wavelength was 270 nm.Results:The concentration range of the four kinds of glycosides were in 12.42-310.50、5.95-148.75、6.85-171.25 and 7.88-197.00 μg · mL,respectively,and recoveries (n=6)were98.6％、104.2％、104.5％、101.0％,respectively,RSD were 1.3％、1.1％、1.3％and 1.6％,respectively.Conclusion:The established RP-HPLC method was simple and fast,which can be used for four flavonoid glycosides of Fengliao Changweikang Jiaonang.
